Associated disorders

The TPM3 gene is associated with autosomal dominant and recessive nemaline myopathy 1 (NEM1) (MedGen UID: 373089) and congenital myopathy with fiber-type disproportion (CFTD) (MedGen UID: 108177).

Pathogenic TPM3 variants probably account for <3% of nemaline myopathy, and may be more commonly associated with CFTD.

TPM3 encodes alpha-tropomyosin which is expressed exclusively in type I (slow) muscle fibers. Tropomyosins bind actin and stabilize the sarcomeric thin filament and regulate muscle contraction.
