PMM2-CDG is the most common congenital disorder of glycosylation, however, due to the overall rarity of these disorders, the percentage of cases attributed to pathogenic variants in PMM2 is unknown.

PMM2 converts mannose 6-phosphate to mannose 1-phosphate in the synthesis of GDP-mannose in the second step of the mannose pathway (PMID: 19862844).
