Pathogenic variants in the MAT1A gene are the only known cause of isolated hypermethioninemia (PMID: 26289392).

The MAT1A gene encodes two isoforms of methionine adenosyltransferase (MAT), MATI (alpha) and MATIII (beta) which function in tetramer and dimer, respectively. MAT catalyzes the formation of adenosylmethionine, an important methyl donor in many transmethylation reactions (PMID: 23150248, 22807109).
