Associated disorders

The ISPD gene is associated with autosomal recessive muscular dystrophy-dystroglycanopathy type A7 (MDDGA7) (MedGen UID: 766244) and type C7 (MDDGC7) (MedGen UID: 807556).

Pathogenic variants in ISPD cause approximately 10% of clinical cases of muscular dystrophy-dystroglycanopathy.

The ISPD gene encodes the isoprenoid synthase domain-containing protein, and is involved in the O-mannosylation of alpha-dystroglycan.

Variants classified as pathogenic or likely pathogenic are confirmed with orthogonal methods, except individual variants that have high quality scores and previously validated in at least ten unrelated samples.

Our analysis detects most intragenic deletions and duplications at single exon resolution. However, in rare situations, single-exon copy number events may not be analyzed due to inherent sequence properties or isolated reduction in data quality.
