Associated disorders

The COL6A1 gene is associated with autosomal dominant and recessive Bethlem myopathy 1 (BTHLM1) (MedGen UID: 331805) and Ullrich congenital muscular dystrophy 1 (UCMD1) (MedGen UID: 98046), collectively known as type VI collagenopathies (MedGen UID: 468393).

Pathogenic variants in COL6A1 are estimated to account for 38% of type VI collagenopathy cases.

Type VI collagen is composed of three chains: alpha1(VI), alpha2(VI), and alpha3(VI). COL6A1, located on chromosome 21q22.3, encodes the alpha1(VI) chain. Collagen VI is found in connective tissue and forms a microfibrillar network, functioning to anchor the basement membrane to the extracellular matrix.

However, in rare situations, single-exon copy number events may not be analyzed due to inherent sequence properties or isolated reduction in data quality.
