Pathogenic variants in the AK2 gene are the only known cause of reticular dysgenesis, which is attributed to an estimated <1% of severe combined immunodeficiency (SCID) (PMID: 8410508, 9063412).
The AK2 gene encodes adenylate kinase 2, which regulates the adenine composition of the cell by catalyzing the reversible transfer of phosphate groups between ATP and AMP. Adenylate kinase 2 is located in the mitochondrial intermembrane space and may be involved in apoptosis (PMID: 26270350).
