Pathogenic variants in the NF2 gene occur in greater than 92% of familial cases and in approximately 70% of simplex cases (i.e., a single occurrence in a family) of NF2.

The NF2 gene encodes merlin, also known as schwannomin, a protein involved in controlling the shape of cells and movement and communication between them. The NF2 gene also functions as a tumor-suppressor gene by inhibiting cell proliferation.
