Pathogenic variants in the GARS gene cause an estimated 3% of clinical cases of CMT and an unknown percentage of dHMN cases.

The gene GARS encodes the protein glycyl-tRNA synthetase. This enzyme is expressed in all cell types and functions in protein synthesis by catalyzing the addition of glycine to its cognate tRNA molecule.
